![]() ![]() To me, it looks like the solver is drawing the line of best fit and then calculating the parameters and using these in the calculations of the predicted values for each standard. The issue that I am running into is when the solver compares the predicted values to the average of the two optical densities for each of the calibration standards' replicates. Basically, what I need to do is walk through each step that the solver performs and compare it to manual calculations. We are trying to validate the solver, but unfortunately it was designed by an outside agency. Since we are a company that produces ELISA tests, we use 4PL for the data. We have an Excel solver that computes the concentration of unknowns based on a calibration curve.
